    New research at the Hebrew University of Jerusalem sheds light on pluripotency?the ability of embryonic stem cells to renew themselves indefinitely and to differentiate into all types of mature cells. Solving this problem, which is a major challenge in modern biology, could expedite the use of embryonic stem cells in cell therapy and regenerative medicine. If scientists can replicate the mechanisms that make pluripotency possible, they could create cells in the laboratory which could be implanted in humans to cure diseases characterized by cell death, such as Alzheimer's, Parkinson's, diabetes and other degenerative diseases.

    To shed light on these processes, researchers in the lab of Dr. Eran Meshorer, in the Department of Genetics at the Hebrew University's Alexander Silberman Institute of Life Sciences, are combining molecular, microscopic and genomic approaches. Meshorer's team is focusing on epigenetic pathways?which cause biological changes without a corresponding change in the DNA sequence?that are specific to embryonic stem cells.

    The molecular basis for epigenetic mechanisms is chromatin, which is comprised of a cell's DNA and structural and regulatory proteins. In groundbreaking research performed by Shai Melcer, a PhD student in the Meshorer lab, the mechanisms which support an "open" chromatin conformation in embryonic stem cells were examined. The researchers found that chromatin is less condensed in embryonic stem cells, allowing them the flexibility or "functional plasticity" to turn into any kind of cell.

    A distinct pattern of chemical modifications of chromatin structural proteins (referred to as the acetylation and methylation of histones) enables a looser chromatin configuration in embryonic stem cells. During the early stages of differentiation, this pattern changes to facilitate chromatin compaction.

    But even more interestingly, the authors found that a nuclear lamina protein, lamin A, is also a part of the secret. In all differentiated cell types, lamin A binds compacted domains of chromatin and anchors them to the cell's nuclear envelope. Lamin A is absent from embryonic stem cells and this may enable the freer, more dynamic chromatin state in the cell nucleus. The authors believe that chromatin plasticity is tantamount to functional plasticity since chromatin is made up of DNA that includes all genes and codes for all proteins in any living cell. Understanding the mechanisms that regulate chromatin function will enable intelligent manipulations of embryonic stem cells in the future.

    "If we can apply this new understanding about the mechanisms that give embryonic stem cells their plasticity, then we can increase or decrease the dynamics of the proteins that bind DNA and thereby increase or decrease the cells' differentiation potential," concludes Dr. Meshorer. "This could expedite the use of embryonic stem cells in cell therapy and regenerative medicine, by enabling the creation of cells in the laboratory which could be implanted in humans to cure diseases characterized by cell death, such as Alzheimer's, Parkinson's, diabetes and other degenerative diseases."

    The research was funded by grants from the European Union (ERC, Marie Curie), Israel Science Foundation, Ministry of Science, Ministry of Health, The National Institute for Psychobiology, Israel Cancer Research Foundation (ICRF), Abisch-Frenkel Foundation and Human Frontiers Science Program (HFSP).

    The research appears in the journal Nature Communications as Melcer et al., Histone modifications and lamin A regulate chromatin protein dynamics in early embryonic stem cell differentiation.

    Eusebio leaves hospital after treatment for stroke

    LISBON, Portugal (AP) -A Lisbon hospital has discharged Eusebio after treating the former Portuguese football great for a stroke.

    The 70-year-old Eusebio spent almost three weeks in hospital after he fell ill at the European Championship last month. After a week in a Polish hospital, he was transferred to Lisbon.

    The Hospital da Luz in the Portuguese capital provided no details of Eusebio's condition, saying he had requested discretion. Eusebio made no comment.

    Eusebio has been hospitalized four times since December with a variety of ailments.

    Eusebio da Silva Ferreira, nicknamed "The Black Panther," became a Portuguese national icon after standout performances in the 1960s and was named by FIFA in 1998 as one of the 10 best football players of all time.

    Survey results by the accountancy KPMG Peat Marwick seems to have gone viral. Nearly half of the respondents to the question of where will the global innovation center of gravity shift in the future answered China. A PC Report article on the matter said, ?44 % of those respondents who believe the center will shift will move to China.?

    I call this the Thomas Friedman Effect: ?Look at all the engineers China is pumping out of its universities. Look at all the R&D centers that are sprouting up in the country. Look at those math test scores, ad nauseam.?

    Egidio Zarrella, of KPMG?s Clients and Innovation Consulting group ? as innovative and confusing a group name as any ? is quoted in the article as saying, ?The survey findings demonstrate that China?s innovation investment has fostered an environment for the development of disruptive technologies that?s growing by leaps and bounds.?

    No it doesn?t. But it?s good PR.

    Closer examination ? that is, living and working in China ? show that most of the engineers that have graduated university have only the most abstract and theoretical notions of the subjects in which they?ve received their degrees due to the stultified education system; and that standardized testing in China does not reflect the degree to which test-takers are able to solve real-world problems with creative approaches in dynamic, heterogeneous conditions; and the companies setting up R&D centers are multinationals here to? localize their product lines for the domestic markets and are loathe to? expose their most precious IP to the Chinese elements.

    Man accidentally eats steel bristle during BBQ

    (AP) ? A Tacoma man needed emergency surgery after accidentally eating a steel bristle from a grill brush he used during a barbeque.

    KING 5 reports that (http://is.gd/J7Tivj ) Adam Wojtanowicz (woh-TAN-oh-witz) went to the hospital Sunday complaining of abdominal pain that wouldn't go away despite his taking medication. Doctors found a metal bristle on a CT scan and performed surgery to get it out.

    Wojtanowicz says he recently hosted a cookout, and he thinks a steel bristle from his grill brush fell onto his steak. He says he apparently swallowed the metal without realizing it.

    The Centers for Disease Control says it knows of at least six people who have recently suffered similar, potentially fatal, injuries. It doesn't blame a particular brand or type of brush.

    Wojtanowicz is expected to recover.

The hard part will be getting your lender to allow you to do the short sale, a lender can refuse to allow you to do a short sale, as stated before this is the tricky part getting the approval when you have not missed a payment and a short sale transaction typically takes months to close.

    You will only have to wait 3 years after doing a short sale if you had to miss payments on your mortgage loan.It may not make a difference how you lose your home either in a foreclosure or a short sale.? The 3 year waiting period for either a short sale or a foreclosure starts from the date of the closing of the transacion. So after those 3 years expire you can put down payment on a place using an FHA loan with only 3.5% along with everyone else buying a home.
    You will see differences in other loan programs that are not FHA loans. loan, and also applying to do a conventional loan.Currently the two most influential investors in the mortgage market are Fannie Mae and Freddie Mac.? These two big govt. sponsored corporations are currently the main investors of the mortgage market for the loans that the banks are doing at this time.To get another home loan you will only have to wait 2 years from the date of closing out on your short sale.? But please keep in mind if you are on another loan whether it is investment property or you co-signed on it, there can be no mortgage lates within the 12 months of application.?? If you foreclose then the waiting period is 5years from the date of your foreclosure date, for a loan that is not a FHA loan.When you become a home owenr again that is a huge difference.? It is going to depend on credit what you have showing???

    There are millons of homeowners that have lost and are losing thier homes and some have even started the process of starting again and becoming a home owner with half of what they owed with the same comparable home they had lost.Homes are seriously cheaper now than they have been and more than likely will not be moving up in value too much in the next 2-3 years.?? In closing if you find yourself in a position of possibly having to do a short sale it can actually be a blessing in disguise.

    Slower Chinese inflation opens door to more easing

    NEW YORK (CNNMoney) -- Prices are tumbling in China, giving the government more leeway to stimulate the world's second-largest economy.

    China's annual inflation rate fell to 2.2% in June, the government's National Bureau of Statistics reported Monday. That's the lowest inflation rate in nearly two-and-a-half years and comes as prices have been falling, especially on food.

    Food prices, which account for more than a third of the inflation calculation, fell 1.6% during the month. Compared to a year earlier, they were up 3.8%.

    Transportation costs, which count for about a tenth of spending, also fell, as did prices for residential water and electricity.

    Overall, China's economic growth has started to slow amid weaker global demand. Simultaneously, inflation has slowed dramatically, freeing China's central bank to stimulate the economy.

    The People's Bank of China has lowered interest rates twice in the last month. The central bank has also tried to spur growth by cutting the amount of money banks are required to hold in reserves.

    Some economists have raised fears of a so-called hard landing in China, in which economic growth slows sharply. Others believe the government still has plenty of room to act, especially now that rapid inflation is not a problem.

    "Inflation continued to ease at a pace slightly faster than expected, which leaves sufficient room for Beijing to take further easing actions," Sun Junwei, HSBC's China economist, said in a note.

    Last month, Fitch Ratings researchers predicted the Chinese economy will grow 8% this year, well below the average of 10.5% per year between 2007 and 2011.

    Even so, Fitch said China's current slowdown does not compare with its much sharper slump in late 2008. ?To top of page

    New Orleans Saints coach Sean Payton files for divorce

    Suspended New Orleans coach Sean Payton has been hounded by bad luck year-around. First Sean Payton was suspended by the league for a year owing to his involvement in the bounty program and now he has to face the ugliness of a divorce with his 20-years wife Beth Payton. To further add salt to the wounds, Sean Payton is just a few days shy of his 20th anniversary.

    The divorce papers filed in the Tarrant County, Texas, were written to protect the couple?s identity. Even though the petitioner and respondent were mentioned through their initials, Sean Payton (SAP) and Beth Payton (BAP), still their identity could be guessed through the couple?s date of marriage and their two children?s birthdays as listed in the records. Furthermore, the divorce records were filed in the system under the title ?Payton.? The filing of the divorce records occurred on 14 June.

    The petition cited the basis for the divorce as the ?discord or conflict of personalities between Petitioner and Respondent that destroys the legitimate ends of the marriage relationship and prevents any reasonable expectation of reconciliation.?

    The legal process of the divorce filing pointed to the Sean Payton?s departure from his home in the suburban Dallas, Texas, where he was living with his wife and two children, 15-years-old Megan and 12-years-old Connor.

    The family moved to the suburban Dallas residence in February, while Sean Payton followed up on his head coach duties with the New Orleans Saints.

    Sean Payton held a 67-37 record with the New Orleans Saints in his six seasons with the franchise and led them to the playoffs three times in a row, including a 2009 Super Bowl XLIV appearance where the New Orleans Saints crushed the Indianapolis Colts with a 31-17 victory.
    The initial petition asked that Sean Payton and his wife be appointed joint custody of their two children while financial issues regarding their estate will be settled without court intervention.
    As seen in most divorces, Sean Payton wife followed-up with her own petition on June 26, maintaining that she should be appointed sole custody of the children and will have ?the exclusive right to designate the primary residence of the children in Tarrant County, Texas.?
    However, Beth Payton agreed any court appointed visits with the children given to Sean Payton, ?as necessary to protect the health and well-being of the children.?

    Beth Payton also demands that she be appointed the sole decision-maker with regards to the children?s legal, medical and educational matters. She also requested that Sean Payton should be held accountable for paying child support to satisfy the children?s? medical and living expenses.

    The path Sea Payton?s personal life has taken will surely be a disappointment for the 48-year-old, who revealed in light of his suspension in April that he will be spending more with his family and son. Sean Payton revealed that his disappointment at not being with the New Orleans Saints next season will b somewhat reduced since he is spending more time with the couple?s two children.

    "Oftentimes with what we do, the time we get to spend with our children is limited," Sean Paton reportedly said then. "So I'm looking forward to taking advantage of that."

    HIV tests offered at pharmacies under pilot initiative

    The US Centers for Disease Control is considering the value of offering HIV tests at US pharmacies, to accelerate identification and treatment of Americans who don't know they are infected with the virus.

    Some 1.1 million people in the US carry HIV, but an estimated 200,000 of them are unaware. The earlier these carriers are identified, the sooner they can begin receiving treatment that prevents their condition from progressing to AIDS ? and the fewer people they will infect unintentionally. At the moment, one-third of those with HIV are diagnosed so late that they develop AIDS within a year.

    "People knowing their HIV status is empowering," says Paul Weidle of the CDC in Atlanta, Georgia, head of the two-year pilot project to offer testing in 12 rural and 12 urban regions with abnormally high burdens of HIV. "If they're negative, that's reassuring in itself, and if they're positive the treatments available are very effective, and they can take steps to avoid infecting others," he says.

    The CDC first introduced routine testing in 2006, but mainly in the context of hospital and family doctor visits. Making the tests available in pharmacies and retail clinics would massively broaden availability because millions of people visit them weekly, and 30 per cent live within a 10-minute drive of a retail clinic.

    Another goal of making testing more available is to limit the stigma of being HIV-positive. "Our goal is to make HIV testing as routine as a blood pressure check," says Weidle.

    Once the pilot finishes, Weidle and his colleagues aim to develop HIV testing "toolkits" that could rapidly be made available to any pharmacies or retail clinics on request.

    In May, the US Food and Drug Administration opened a new front against the disease by recommending that antiviral drugs be offered to people who don't carry HIV but are at risk of infection, perhaps due to their lifestyle or because their partners are infected.

    The move is symptomatic of a global shift towards prevention. As part of that, the WHO is also keen for tests to be offered more widely elsewhere in the world.

    Cell biology: New insights into the life of microtubules

    ScienceDaily (July 2, 2012) ? Every second, around 25 million cell divisions take place in our bodies. This process is driven by microtubule filaments which continually grow and shrink. A new study shows how so-called motor proteins in the cytosol can control their dynamics.

    The cytoskeleton plays a central role in the process of cell division. It is composed in large part of protein filaments known as microtubules, which also help determine the size, shape and mobility of a cell. In a new study, LMU biophysicist Erwin Frey and his colleagues Anna Melbinger and Louis Reese have used a theoretical model to show how cells control the construction and breakdown of microtubules. The dy-namics of this process affect how cells divide, and how they maintain the cytoskeleton. In particular, it is responsible for regulating the size and shape of the mitotic spindle.

    Easy come, easy go

    Theoretical modeling has now revealed that the regulation of microtubule length relies on the length of the filament itself: The longer the filament the more motor proteins can attach to it. These all move towards the 'plus end' of the microtubule and tend to pile up as they do so. Upon arrival at the plus-end they shorten the filament. In parallel, new microtubule building blocks bind to precisely the same 'plus end' through spontaneous polymerization from the surrounding cytosol, and the filament grows.

    It has now been demonstrated that such interplay between growth and length-dependent shrinkage indeed results in the maintenance of a precisely regulated microtubule length. This kind of length regulation might be essential for many intracellular tasks which depend on microtubules of a certain length.
